Sensitive analysis of bioactive peptides with two functional groups derivatization-liquid chromatography-tandem mass spectrometry

We have developed a novel method for sensitive analysis of bioactive peptides in biological sample using two functional groups derivatization-liquid chromatography-tandem mass spectrometry. In this study, angiotensin I, II, III, IV, bradykinin, nerotensin and neurokinin A were used as model compounds. At first, amino group of bioactive peptides was derivatized with acetaldehyde-d4 or acetaldehyde for obtained different mass. Because this derivatization reaction was simple and quick, and derivatization reagent was easy to remove in evaporation, high-yield different mass bioactive peptides were obtained and accurate comparative analysis has been achieved. Next, carboxyl group of bioactive peptides was derivatized with 1-(2-pyrimidinyl)piperazine for sensitive LC-MS/MS analysis. About 10-fold sensitive analysis was achieved by this derivatization.
