Investigation of Relationship between Conformation and Activity of Biopharmaceutical Using Hydrogen/Deuterium Exchange Mass Spectrometry

Information on the higher-order structure (HOS) is important in biopharmaceutical development. Recently, hydrogen/deuterium exchange coupled with mass spectrometry (HDX-MS) has been a useful tool to evaluate protein conformation, and automated HDX-MS systems are now available in commercial. To investigate the relationship between conformation and activity of biopharmaceuticals, degraded samples were subjected for characterization of physiochemical property, biological activity, and HOS. As a result, we could demonstrate that HDX-MS can provide the information on the extent and location of the structural damage.
