Analytical Characterization of Antibody-Drug Conjugates via Mass Spectrometry

Mass spectrometry is an important tool in the development of biopharmaceuticals. Elucidation of structure by liquid chromatography-mass spectrometry (LC-MS) yields important information such as post-translation modifications on proteins. In more complex biologics, such as antibody-drug conjugates (ADCs) that have been subjected to chemical modification by conjugation of small molecules, mass spectrometry is used to identify conjugation number, conjugation sites and the conjugation distribution profile.1 A case study will be presented showing the characterization of ADCs via peptide mass mapping, intact mass analysis and size-exclusion chromatography coupled to a mass spectrometer (MS-SEC).
