Development of High Throughput Quantitative Method of Oligonucleotide Therapeutics by Using LC/MS/MS

Oligonucleotide therapeutics such as antisense are expected as the next generation drugs which interfere with synthesis of target proteins by hybridization with their specific mRNA. Quantitative method of oligonucleotide in biological matrices using LC/MS is useful for drug discovery and development. However, there are only a few reports on LC/MS methods of oligonucleotide and many issues such as throughput of sample preparation or control of chromatographic behavior need to be solved. We developed a high throughput quantitative LC/MS/MS method by optimizing the MS detection, HPLC method, and the procedure of sample preparation.
