The usefulness of differential mobility spectrometry (DMS) device, SelexION, coupled with 6500QTRAP (AB SCIEX) for drug discovery such as cassette dosing study is demonstrated using structural analogs with close molecular weights. Optimizing compensation voltage (CV) and appropriate parameters for DMS with good sensitivity and selectivity is key factor to succeed in developing the bioanalytical method. The performance of the bioanalytical method using SelexION to quantitate three analytes in rat plasma, with three different CV values, is assessed in terms of linearity of standard curves and accuracy and precision of quality control samples without any internal standards. In addition, we would introduce the advantage of application of SelexION and 6500QTRAP to develop bioanalytical method of middle weight molecules such as peptides and antisense -oligonuclueotides.
