The use of a non-volatile solution such as phosphate buffer or ion pair reagent solution as the mobile phase in LC analysis is an effective means to perform the retention and separation of the target components. However, it is well-known when connected to a mass spectrometer, it inhibits the ionization of the target components and causes contamination of the interface. Therefore, although the reexamination of the mobile phase conditions is required, in particular when ion pair reagent is used, it is difficult and time consuming.
A proposed LC-SPE method, in which solid phase extraction cartridges are located downstream of the separation column, allows us to isolate/fractionate specific components contained in the sample. We successfully applied this method to the measurement of mass spectra of target component without changing the ion pair reagent solution as mobile phase.
