Easy and Fast Method for Quantitative Determination of Long-Chain Fatty Acyl-CoAs

Long-chain fatty acyl-CoAs (LCAcyl-CoAs) are key substrates for lipid biosynthesis and oxidation, and are concerned with many important enzyme reactions in lipid metabolism. LCAcyl-CoAs are very important as markers of fatty acid metabolic pathway. However, a setup of their quantitative analytical method using LC/MS/MS is very difficult; because LCAcyl-CoAs have phosphate groups generally known to cause a tailing and adsorption, and because it is required to separate LCAcyl-CoAs which have close molecular weights by HPLC.
We performed examination of sample preparation method and HPLC conditions in order to develop an easy and fast method for the quantitation of LCAcyl-CoAs (C16:1, C16:0, C18:3, C18:2, C18:1, C18:0) in mouse liver by LC/MS/MS. As a result, we established an analytical method for LCAcyl-CoAs in only 3 minutes with a simple deproteinization.
