Method Development for Quantification of Antibody Drug using LC/MS/MS to Measure of Antibody Drug in Mice Serum
The method for quantification of antibody drug (anti her2 antibody) in mouse serums with LC/MS/MS was developed. Endogenous IgG including anti her2 antibody was extracted from the serum using protein G and digested with trypsin. Anti her2 antibody-specific peptides were selected from amino acid sequence acquired through a MASCOT search, and the most optimum peptide was quantified selectively as a target ion. LC/MS/MS analysis was conducted using serums collected over time from mice with single intravenous administration of antibody drug. As a result, concentration profile resulting from the measured concentrations was almost comparable to ELISA results stated in the application. The method would be applicable for quantification of an antibody drug in biological samples.
