Quantitative proteome analysis of central metabolic pathway enzymes by nanoLC-MRM-based platform using ultra-fast mass spectrometry

A liquid chromatography-multiple reaction monitoring (LC-MRM) technique was developed by taking advantage of the fast scanning speed of ultra-fast mass spectrometry (UFMS) for the widely targeted analysis of enzymes related to central metabolism. A set of LC-MRM conditions of 4000QTRAP for the enzyme analysis of yeast central metabolism was transferred to LCMS-8040. Based on this, a method for the widely targeted analysis of more than 200 peptides was successfully developed. The enzyme levels in the single gene disrupted strains were successfully compared by using yeast cells cultured in 13C and non-labeled media. The results demonstrated that UFMS-based LC-MRM is a promising tool for system-level understanding of the regulation network of the central metabolic pathway
