新規 caged クックソン型試薬を用いた実用的ビタミンD誘導体化およびLC-MS/MS定量法の開発

Introduction It is important to quantify accurately serum 25-Hydroxylated vitamin D【25(OH)D】because it is used for evaluation of bone health, and also because 25(OH)D and its metabolites are reportedly responsible for a risk of various diseases. LC-MS/MS is necessary to accurately measure vitamin D metabolites and a Cookson-type reagent, 1,2,4-triazoline-3,5-dione(TAD) is often used to improve their ionization efficiency. For routine use in clinical laboratories, it is desirable that derivatization reagent is user-friendly and stable during long-term storage, in addition to high efficiency in ionization. For these reasons, we have developed a novel caged Cookson-type reagent, (14-(4-(dimethylamino)phenyl)-9-phenyl-9,10-dihydro-9,10-[1,2]epitriazoloanthracene-13,15-dione, DAP-PA). We here report LC-MS/MS-based method for simultaneous measurements of human serum 25(OH)D3, 3-epi-25(OH)D3, 25(OH)D2, and 24,25(OH)2D3 in clinical laboratories using DAP-PA as a derivatization reagent.
