ノンターゲットリピドミクスの現状と課題

Lipid metabolites consist of a wide variety of fatty acids or of polar groups, and some of them are quantitatively and qualitatively variable or structurally converted by indigenous bacteria. To investigate lipid metabolic patterns and changes in detail, non-targeted lipidomics analysis by high-resolution LC-MS system has been wildly applied. However, it has some technical issues in the conventional approaches regarding detection sensitivity and identification accuracy. To solve these problems, we present a multi-lipidomics approach which combines non-targeted method using LC/Q-TOF MS and Lipidiscovery (in-house screening software), and targeted method using LC/TQ MS in accordance with lipid properties and the concentration range. Also, we introduce a high-precision structural analysis approach not only by collision-induced dissociation (CID) but also by electron capture dissociation (ECD) which has a strong potential that helps to specify the positions of hydroxyl group and double bond or to identify unknown lipid molecular species.
