The Analysis of Protein Structure by Mass Spectrometry

Two biological applications of mass spectrometry method onto protein structural analysis will be illustrated in this talk. The first topic will be focusing on the protein spatial structure which would be examined by chemical cross-linking method coupled with mass spectrometric analysis. A novel data analysis algorithm has developed for the analysis of peptides with chemical cross-linkages. The second topic is to identify alternative splicing forms of proteins, a biological process to produce multiple proteins from one single gene, by mass spectrometry technique. Unlike traditional transcriptomics-based technique, we have developed a generic method to confirm the existence of alternative splicing proteins by the typical shotgun proteomics experiment. These two biological applications have demonstrated that mass spectrometry is a key technique and provide irreplaceable information for protein structural analysis.
