LA-ICP-TOF-MSを用いた生体組織内ナノ粒子・元素の同時イメージング分析

With the increasing use of NPs, the potential risk of NPs for human health is increasingly concerned. Recent studies revealed that the size of NPs is also important to control the toxicity of the metal NPs. Moreover, changes in elemental distributions of several key biologically relevant elements (e.g., Mg, Fe, Cu, or Zn) would be another important parameter to evaluate the toxicity of the metal NPs. Faced with this, we have developed new analytical technique to define the position, size of the NPs, and elemental distributions of key nutrients using laser ablation-inductively coupled plasma-time-of-flight mass spectrometry (LA-ICP-TOF-MS). In this study, imaging analyses of NPs and biologically relevant elements were conducted on liver tissues from mice. To define the size of the NPs, calibration standard using 60 nm Ag NPs dispersed in a photocurable resin was prepared. The imaging technique, together with the size calibration using our in-house standard can provide key information to understand the transport and toxicity of the metal NPs.
