高分解能質量分析計を用いた標的SIM法による腫瘍マーカーの定量解析
In targeted proteomics, MS-based assay provides a quantitative, sensitive, and reproducible approach for analysis of proteins in any biological samples. Selected/multiple reaction monitoring (SRM/MRM) using triple-quadrupole instrument is the most widely used for the quantification of specific proteins/peptides, although it is still challenging to quantitate low abundant proteins/peptides in complex samples from clinical specimens. In this study, we investigated the establishment of targeted selected ion monitoring (t-SIM) methodology using nano-liquid chromatography system coupled with a Q-Exactive high-resolution accurate mass spectrometer. Furthermore, we applied the well-established t-SIM platform to relative quantitation of neuron-specific enolase (NSE, gamma-enolase), one of small-cell lung cancer (SCLC) biomarkers, in cancerous cells laser-microdissected from formalin-fixed paraffin-embedded tissue samples from SCLC patients.
