LC/IMS/MSにおける前駆体イオン情報のみを用いるプロテオーム解析法の開発

In proteomics LC/MS/MS systems, high efficiency in both LC and MS separation is essential because of the high complexity and the wide dynamic range of the digested peptides from proteome samples. Recent development of ion mobility spectrometry coupled with LC/MS (LC/IMS/MS) provides additional separation dimension for peptide separation, resulting in remarkable improvement in the peak detection. However, it is still a challenging task to identify the substantial fraction of the detected but unidentified peaks because the quality of the product ion spectra is not high enough even when the precursor ions are detected. Here, we developed a novel approach in which peptides are identified based on only the precursor ion information such as m/z, retention time and ion mobility in combination with the library-matching approaches.
