ToF-SIMSを用いた皮膚組織中低分子化合物のイメージング質量分析
Retention of water in human skin is important. This function is performed by low molecular weight compounds in the stratum corneum. To understand its mechanism, it is helpful to clarify the distribution of these compounds in the stratum corneum. There are some methods to examine it, and imaging mass spectrometry (IMS) is one of these methods. Especially secondary ion mass spectrometry (SIMS) has high spatial resolution. This study aimed at the establishment of the analysis method on the distribution of low molecular weight compounds including amino acids in stratum corneum with high spatial resolution. Skin samples were prepared by tape stripping. 20 tapes were taken from the same position to remove the stratum corneum, and the 2nd and 20th tapes were analyzed to investigate their depth distribution. Furthermore, the standard samples of the three amino acids (Arginine, Lysine, and Serine) were prepared and analyzed to clarify the principal peaks derived from these amino acids. Then, the distribution of these three amino acids in the stratum corneum was investigated using these peaks.
