A Facile Method to Characterize the Double Bonds of Unsaturated Fatty Acids by Matrix-Assisted Laser Desorption Ionization Mass Spectrometry

In this study, we examined our previous method and found it useful for rapid identification of unsaturated double bonds in fatty acids.
A hydrophilic peptide, “Bradykinin” was selected as a carrier of fatty acid, named here “fatty acid-linked peptide”(FALP), which made the handling and measurement of fatty acid molecules easier.
An aliquot of the resultant FALP solution (0.1%TFA H2O-acetonitrile, was blotted on a standard stainless-steel plate, dried up, and exposed to ambient air for 1~3 hours; thereafter, each spot was mixed with matrix and measured by MALDI-MS. FALP was observed at m/z 1296.6 while the FALP that was kept under the ambient air for 3h distinctly shifted to m/z 1214.6. This molecular mass could be readily assigned to the degradation product (aldehyde form) as a result of oxidative cleavage of the double bond at position 9. The results of other kinds of unsaturated fatty acids also showed cleavage at position of their olefinic bonds.
