Evaluation of DIUTHAME for High Throughput Screening
The performance characteristics of desorption ionization using through hole alumina membrane (DIUTHAME) are thought to be suited for applications relying on automation, such as high throughput screening (HTS). This fact was confirmed by monitoring protein functional activity shown in DIUTHAME spectra. Acetylcholine esterase (AChE) enzyme reaction assays were performed then analyzed by using DIUTHAME. The interaction of the enzyme with the substrate molecule was successfully recorded as peak intensities of the substrate acetylcholine (ACh) and the reaction product choline (Ch) varying in accordance with the reaction time course. Besides, the analysis using DIUTHAME indicated high tolerance for additives such as tris(hydroxymethyl)aminomethane hydrochloride (Tris-Cl) or dimethyl sulfoxide (DMSO).
