Identification and Subsequent Relative Quantitation of Stable Isotope Labeled Released N-Glycans by LC-, ESI-, Tandem MS with SimGlycan software

Glycans play a critical quality attribute for the therapeutic monoclonal antibodies, where terminal sugars of the Fc glycans are responsible for determining their efficacy and safety. Glycomics analysis involves correct identification of the glycans as well as their subsequent quantitation in the samples. Mass spectrometry is one of the most popular analytical platform in glycomics research. This technology facilitates not only the identification of the monosaccharide compositions of glycans using the information provided by the glycosidic fragment ions, but also detailed investigation of their topological structures using information provided by the cross ring fragment ions. Stable isotope labels facilitate accurate identification, and subsequent quantification of glycans using LC-MS or ESI-MS workflows. Major types of stable isotope labeling approaches reported in literature includes (1) mass shift, and (2) isobaric tags. Efficient software tools are required to exploit the recently developed above-mentioned glycomics methods. We have developed SimGlycan software modules to streamline the analysis of data generated by LC-, ESI-, MS based qualitative and quantitative methods.
