Gel-enhanced Approach for Quantification of Low-abundance and Low-molecular-weight Cargo Contents in Serum-derived Exosomes

Much attention has been paid to exosomes as non-invasive diagnostic tools, namely “Liquid Biopsy”, and as an ideal source for the discovery of new biomarkers. In particular, serum- and plasma-derived exosomal cargo contents (luminal proteins) are of interest as stable biomarkers for clinical diagnosis in cancer and neurodegenerative diseases. Here, we will describe the gel electrophoresis-based enrichment of low-abundance and low-molecular-weight cargo contents (e.g. cytokines) in a 10-20 kDa protein fraction prior to quantitative LC-MS assay. It is expected that this gel-based enrichment approach can reduce the background proteome complexities in serum contaminants and exosomal major proteins; thereby leading to increase the chances of detecting minor cargo contents associated with development and progression of cancer and other diseases. To evaluate the capability of this approach, we have tried to detect and quantify exosomal macrophage migration inhibitory factor (Exo-MIF+), which is a prognostic marker for pancreatic ductal adenocarcinoma (PDAC) and liver metastasis, in serum from normal controls and patients with PDAC. In this presentation, the usefulness of our approach is discussed in more detail.
