Middle down analysis for histone H3 modification
Histones are highly modified molecules and their modifications are thought to be involved in epigenetic regulations and memory. Using Orbitrap Fusion, we have acquired high-resolution ETD spectra of histone H3.1 tail aa 1-49 peptides. We have found that the spectra have two main characteristics. First, the spectra had mixed ones of the positional isomers of modifications. Second, ETD for long peptides shows high frequencies of hydrogen rearrangements to provide 1-Da-gapped-clusters of ions. We are now doing efforts to resolve the mixed components and to improve charge determination using the hydrogen-rearranged clusters of ions.
