Development and application of strategy for comparative analysis of protein expression and functional state.

Recent technological advances in shotgun proteomics have made human proteome catalogues available to the scientific community. However, it is difficult to obtain information of cleavage and interaction of proteins. On the other hand, the method referred to as the GeLC-MS/MS based on electrophoresis is examine functional status of protein such as proteolytic events, protein interaction, and post translational modification. In this study, we developed the method that combine GeLC-MS/MS with stable isotope labeling of proteins prior to electrophoresis. It is possible to comparatively analyze functional status of proteins in detail. Here, we applied this strategy to analyze alternation of protein expression and function in mouse hippocampus induced by water immersion restraint stress.
