Nanoparticle Assisted Laser Desorption/Ionization for Imaging Mass Spectrometry of Biological Samples

Recently, Surface-assisted laser desorption/ionization mass spectrometry (SALDI-MS) using nanoparticles has been used for the LDI-MS analysis of small molecules as an organic MALDI matrix-free approach. Compared to MALDI-MS, SALDI-MS affords certain advantages, such as easy sample preparation, low background noise, better reproducibility. Also, the use of an organic matrix in MALDI Imaging MS can limit the spatial resolution because of the large matrix crystal size formed by MALDI matrix. We have analyzed two standard lipid samples cholesterol, (DSPC) and also the lipids (extracted from amoeba cells), intact amoeba cells using various nanoparticles such as TiO2, CeO2, WO3, Ag, Pt, ZnO, CaO and also with DHB matrix. The cholesterol has been successfully analyzed using all the NPs and DHB matrix whereas DSPC was successfully analyzed by WO3, CeO2 NPs, and DHB matrix only. The lipids extracted with amoeba cells and intact amoeba cells have been successfully analyzed with only DHB matrix. In future, we are planning to image a single intact cell by SALDI MS using nanoparticles or nanostructured surfaces. Since single cell analysis is very useful in knowing cellular functions etc., this technique would be very useful in future for single cell imaging.
