Evaluation of Novel Administration Method Using Imaging Mass Spectrometry for Dynamic Analysis of Transdermal Administered Drugs

Transdermal vaccines are considered to be the promised new vaccine administration method. However, there are no adjuvants which enhance the effectiveness of vaccines and are administrated with vaccines by transcutaneous administration. Therefore, it is necessary to observe the distributions of new adjuvants in the skin tissue. The purpose of my study is investigation of the adjuvants administrated by transcutaneous in the skin tissue. In this research, I evaluated the difference between the two methods of transdermal administration. One method is to attach a patch loaded with imiquimod, an adjuvant, to skin tissue for 24 hours. Another method is to form a puncture hole in the skin tissue using a microneedle, drop imiquimod onto it, and then apply the patch to the surface for 6 hours. Dynamic analysis of imiquimod was performed by using imaging mass spectrometry of samples prepared by these two methods. As a result, [imiquimod+H]+ was observed from the vicinity of the skin tissue surface in the method of applying the patch loaded with the drug on the skin tissue surface, but in the other method, [imiquimod+H]+ was observed from the entire sample.
