Accuracy of Peptide Isotope Distribution in Mass Spectra

Isotope distribution of peptide peaks in mass spectra can be used to support the peptide identification, although it has not been so frequently used in proteomics. In this study, we examined the accuracy of the observed isotope distribution of tryptic peptides. We evaluated several factors to influence the similarity between the observed and theoretical isotope distribution. As a result, it was found that the absolute intensity as well as the charge states were the two major factors to affect the accuracy in measuring the isotope distribution of peptides.
