Identification of sialyl linkage glycan isomers on glycosphingolipid by SALSA-glycoblotting method
Sialic acids exist the nonreducing ends glycans on glcoconjugates with α2,3 and α2,6 linkage. These sialylated glycans are critically involved in many biological events, such as cell adhesion, signal transduction, and virus infection. Glycosphingolipids (GSLs) have a large number of sialyl linked glycan isomers and α2,8 linked polysialic acids. Recently, Nishikaze et al. reported sialic acid linkage-specific alkylamidation (SALSA) method, which allows the discrimination of sialic acid linkage isomers on N-glycans by mass spectrometric analysis. In this presentation, we report that the comprehensive GSL-glycome analysis by SALSA-glycoblotting method combined endoglycoceramidase digestion. Additionally, we introduce the novel method of lactone-opening and amidation with various linear alkyl amines for α2,3 and α2,8 linked sialic acid, which can significantly reduce total reaction time of general SALSA-glycoblotting method.
