Development of SFE-SFC/MS with a novel split-flow system

The online coupling of extraction with chromatography could improve the efficiency and accuracy of the entire analytical process. We have developed a novel split-flow system for online supercritical fluid extraction-supercritical fluid chromatography/mass spectrometry (SFE-SFC/MS) using supercritical carbon dioxide (SCCO2) and modifier as the mobile phase. Previous systems suffered from serious peak distortion and carryover due to high amounts of extracts, since the entire extract would be transferred into the chromatographic column. In our system, the extracts were split and introduced to the column by controlling the pressure of two back pressure regulators (BPRs) located before and after the column. The developed online SFE-SFC/MS with a novel split-flow system was also verified using a pesticide-spiked sample.The extracts was successfully split and introduced to the column at a high split ratio, which improved the peak shape distortion of compounds in the separation section.
