質量分析による抗体医薬と抗原の相互作用解析

Antibody drugs are now effectively used for the treatments of cancer and autoimmune diseases. Therapeutic antibodies form complexes specifically with antigen, so called immune complexes (ICs), which results in blocking of signal transduction among cells and/or invoking immune responses such as complement dependent cellular cytotoxicity and antibody dependent cellular cytotoxicity. In the case of a oligomeric antigen, the antibody to the antigen, that is dimer thus has two binding sites, is expected to form ICs with the antigen under different stoichiometries. Here we show our recent studies on the interaction between an antigen, tumor necrosis factor (TNF), and its monoclonal antibodies (mAbs), adalimumab and infliximab, by native mass spectrometry. In the conference, results from MS will be discussed based on three-dimensional structures of TNF/Fab complexes and on the comparison with results from other biophysical methods.
