MALDIイメージング質量分析法を用いたアルドステロン特異的可視化法の開発

Aldosterone, an adrenocortical hormone, plays an important role in regulation of sodium and water homeostasis. In studies of endocrine disease, aldosterone production in adrenal gland is region of interest, however methods for identifying aldosterone on adrenal sections have not been established. The purpose of our study is to develop the method for visualizing aldosterone directly and specifically using MALDI imaging mass spectrometry (MALDI-IMS). To detect aldosterone specifically in adrenal gland using MALDI-IMS, increase in ionization efficiency and discrimination from structural isomer are required. Therefore, we applied on-tissue derivatization with Girard’s T reagent and MSn analysis using ion-trap mass spectrometer. In this presentation, introduction of the proposed methodology and the application to human clinical samples of adrenal gland will be provided.
