タンデム四重極質量分析計を用いたバイオ医薬品の定量分析の感度向上

Xevo TQ-XS used here is fitted with a novel ion guide which results in increased transmission of ions of interest. This, combined with modifications in the source helped enhance sensitivity for the biotherapeutic molecules tested. An improved detection system has the added advantage of extending the dynamic range of the instrument and is evaluated.
Analyte area counts were compared between Waters Xevo TQ-S and TQ-XS. For Vancomycin, an increase in area counts of >5 fold was observed across the entire analytical range of 100 pg/mL to 10 μg/mL tested. Signal to noise across the same analytical range was also compared between the Xevo TQ-S and TQ-XS. For Vancomycin, an increase in signal to noise of >3 fold was observed across the entire analytical range.
For a monoclonal antibody spiked in plasma and digested using the Waters ProteinWorksTM digest kit, the increase in analyte area counts was >2 fold when compared to Xevo TQ-S system.
Other critical bioanalytical parameters including accuracy, precision and robustness of the system is currently being tested on several candidate molecules and will be discussed.
