糖ペプチド用シアル酸結合様式特異的修飾法の開発

We have recently developed Sialic Acid Linkage-Specific Alkylamidation (SALSA) for linkage isomer discrimination of sialic acid residues on glycans. SALSA converts -COOH groups on sialic acid residues into alkylamide forms with different length of alkyl chains in linkage-specific manner, allowing direct discrimination of sialyl linkages based on the difference in m/z values. This method has been developed for (released) glycan analysis; therefore, SALSA for glycopeptide derivatization is also conceivable as a next step. However, chemical derivatization of glycopeptides is challenging because of the difficulty in suppressing unfavorable side-reactions originated from the presence of various amino acid residues. In this study, we developed a promising workflow to derivatize sialic acids on glycopeptides in a linkage-specific manner, which can be applied to various glycopeptide samples without causing unfavorable side-reactions.
