安定同位体標識誘導体化を利用したLC-HR/MSによる官能基特異的高精度キラルメタボロミクスの開発

Metabolomics, comprehensive analysis of metabolites, has now become an importance in various research areas. Recently, the relationship of enantiomeric metabolites and diseases is attaching attention. However, no comprehensive analysis of these compounds has been tried because of the difficulty of chiral separation and detection from complex matrices such as biological sample.
Based on these backgrounds, we developed a chiral and achiral non-targeted metabolomics using chiral derivatization method by LC-tandem triple quadrupole MS/MS. This method enables highly sensitive and selective detection of carboxylic acids and amines, combined with chiral derivatization and precursor ion scan by MS/MS analysis. On the other hand, some disadvantages were associated in this method. One is the identification difficulty of metabolite by the low mass resolution. Only one determination of characteristic ions of the derivatives is another drawback.
Thus, we propose a novel approach which can obtain high accuracy mass and quantitative data by combination of LC-high resolution mass spectrometry and isotope coded derivatization correction. Of cause, mass accuracy was high enough to data base searching from m/z of derivatives. Furthermore, the method, named isotope corrected chiral and achiral non-targeted metabolomics, enables more accurate and sensitive comparison analysis than previous method.
