プロトメタボリズム研究における生体分子前駆体検出のためのマイクロ固相抽出・誘導体化法

Although glycoaldehyde, glyceraldehyde, ribose, and amino acids are indispensable precursor sets for the proto-metabolism1), the effective formation and accumulation processes of these molecules have been still unclear in the molecular evolution during the Hadean earth. A simple analytical procedure has been demanded to evaluate the formation or accumulation of these small polar to semi-polar compounds in aqueous samples obtained from various model synthesis. GC-MS after TMS and TBDMS modifications is one of the ways to determine the targets using one mass spectrometer setting, however it takes time and effort to conduct complex extraction and derivatization procedures involving solvent extractions. Therefore, we applied solid phase micro extraction (SPME Arrow) and on-fiber derivatization to analyze the small sugars, amino acids. Combination of NIST database search combined with GC-MS 70 eV electron ionization (EI) was advantageous to characterize diverse molecular analogues which have similar structures of the target molecules. Optimal extraction, derivatization, and the introduction in the on-line sample preparation were investigated. Using this technique, formation of sugars and amino acids in the radiolysis were investigated for gamma ray exposure aqueous 0.1mM HCN and aqueous CH3CN(30%).
