ペプチド構造解析における水素付着/脱離解離法(HAD)の有用性
A commonly used fragmentation technique in mass spectrometry is CID for structural characterization of various peptides. However, CID has a disadvantage of low sequence coverage for peptides containing labile PTMs, multiple basic amino acids residues, disulfide bonds and so on. We have developed a new fragmentation technique “HAD (hydrogen attachment/abstraction dissociation)” for gas-phase peptide ions and reported in ASMS 2015, in which HAD showed similar fragmentation of ETD/ECD and provided consecutive c and z-type product ions retaining labile PTMs such as a phosphate group and O-GalNAc. In this study, we applied HAD for various peptides that have low fragment coverage by CID and showed an advantage of HAD for peptide structural analyses.
