プロテオーム間引き解析とSWATH

Together with modern mass spectrometers with high resolving power and high mass accuracy, ultrahigh resolution LC based on meter-long monolithic silica capillary columns have been employed to expand the separation space for shotgun proteome samples, resulting in higher sensitivity and specificity in quantitative proteomics. We explored this approach named ‘one-shot proteomics’ without pre-fractionation to identify as many proteins as possible. However it still remains a challenging task to uncover the entire human proteome. Here, we propose a new strategy called ‘slicing proteomics’, in which the target proteome is sliced at peptide level to reduce the sample complexity and is subjected to one-shot proteomics. In this presentation, I will show some examples to use different approaches for slicing the human proteome including narrow range SWATH acquisition.
