目から鱗のSRM応用:疾患関連タンパク質研究への活用例

Selected reaction monitoring mode (SRM)-based strategies have recently been developed for quantitation of proteins. Using sequence information from the proteins, these strategies can be used to develop simultaneous quantitative methods for several proteins of interest. From a biologist’s viewpoint, an SRM-based strategy, which does not require specific antibodies, would be a convenient method for protein analysis.
In this workshop, I will discuss the problems and potential roles of SRM-based strategies in protein analysis with the panelists and audience. First, I will present a problem with protein quantitation using SRM from our study. Second, I will present how SRM can be applied to post-translational modification profiling of proteins. Finally, I would like to propose a role for SRM in hypothesis-driven biological research. SRM strategies can be used to develop the detection methods for specific proteins using the mass of specific peptide sequences derived from these target proteins. These strategies can be applied to proteins that are predicted by biologists, but not registered in the sequence database. I will introduce a case from our study of spontaneous animal amyloid AA amyloidosis.
I hope to conduct a lively discussion on non-stereotyped applications for SRM in this workshop.
