超臨界流体クロマトグラフィー/質量分析を用いた胆汁酸の一斉分析

Bile acids play an important role in digestion and fat solubilization. Additionally, extensive studies in the past decade have revealed that bile acids function as the signaling molecules to regulate metabolisms. These bile acids controlled signaling pathways have become the source of drug targets for metabolic diseases. Therefore, an analysis method determining bile acid profile is highly sought after. Bile acids are complex compounds because of their wide polarity and structural analogs, resulting from the presence of unconjugated and conjugated forms with varying the number and orientation of hydroxyl group. A number of analysis methods using LC/MS and GC/MS have been reported. However, there is no report for simultaneous bile acids analysis using supercritical fluid chromatography (SFC). In this study, we have successfully developed a method using SFC coupled to ESI/MS/MS for comprehensive bile acid profiling.
As a result, 25 bile acids including conjugates were separated simultaneously within 13 min. The method was applied to rat serum samples, 24 of the bile acids were quantified. This study not only reports simultaneous analysis of bile acids including conjugates but also indicates the method is applicable to a biological sample. This is the first report on the simultaneous analysis of bile acids using SFC/MS. The developed method will be an alternative to existing analysis methodology for studies on the bile acid metabolism in the medical and pharmaceutical fields.
