Cysteine is the one of least abundant amino acids, it is frequently located in a biologically important site in a protein such as a cofactor binding site. In a human protein sequence database, about 97% of proteins and peptides contain cysteine residues. In this context, a method for the selective isolation and detection of proteins/peptides containing free thiol groups may lead to the discovery of novel functional roles of Cys residues in proteins.
