ポリマー系イオンクロマトグラフィー用カラムShodex IC YS-50を用いた各種神経伝達物質のLC/MS測定

Shodex IC YS-50, a poly vinyl alcohol-based ion chromatography column (for cation), was used to develop high-sensitive LC/MS method for the quantification of various neurotransmitters. Acetylcholine is a neurotransmitter that synthesized from choline. Their presence is related to several life phenomena and diseases, and it is important to be able to quantify these molecules. In order to simplify conventional method HPLC-ECD that need a post-separation affinity column modified with choline oxidase and acetylcholine esterase, a new LC/MS method was studied. We have found that Shodex IC YS-50, with nitric acid aqueous solution/acetonitrile as eluent is useful for this analysis. Peak shapes of choline and acetylcholine were sharp with baseline separation, and the calibration curves were linear. Both detection limits were 0.005 μM. Moreover, it have been demonstrated that this method is applicable for various amine or amino acid neurotransmitters. Our described LC/MS method should be more facile with higher selectivity than the previous method.
