A determination of various endogenous metabolites is important to evaluate the efficacy and toxicity of drugs and to investigate the mechanism of action. Among them, steroid hormones play broad and pivotal roles in the regulation of gene expression, reproduction, cardiovascular, and neurological functions. Hence, determination of the steroid hormones in biological sample is sometimes critical in the drug development, and the method validation to measure traces of steroid hormones with easy procedures is required. We have developed a high sensitive analytical method of 12 endogenous steroid hormones in a variety of plasma. This method was conducted for 200 μL of a variety of plasma after liquid-liquid extraction using LC-MS/MS. In this high sensitive analytical method, approximately 10 to 30 pg/mL of lower limit of quantification can be obtained for all the 12 steroid hormones. This method can be used for simultaneous routine measurements of steroid hormones in plasma samples.
