Live Single-cell MSによるダイレクト脂質分析

“Live Single-cell Mass Spectrometry” traps cellular components of a cell, and provides us direct and real-time molecular analyses of cell behavior-linked molecular profile. In this study, we applied this method to trace metabolism of rat basophilic leukemia (RBL-2H3) as an allergy model cell line under stimulation of calcium ionophore. From the results of a single RBL-2H3 cell lipid analysis, more than 2,000 peaks were detected by positive and negative ion modes. Most of the peaks which detected by positive ion mode were identified phosphatidylcholines and sphingomyelines. Other phospholipids, phosphatidic acids, phosphatidylethanolamines, phosphatidylserines and phosphatidylinositols peaks were detected by negative ion mode. Furthermore, about 200 peaks were listed as significantly-increased/ decreased peaks after calcium ionophore stimulation. These results show that directly and real-time allergy reaction-linked lipid metabolism was analyzed by a Live Single-cell Mass Spectrometry.
