Advanced-nano-LC/MALDI-MSを用いた血漿中の糖タンパク質E-カドヘリンの糖ペプチド分析

MALDI-MS is a powerful tool for proteomics and glycomics so on. In MALDI-MS analysis for biological samples, “Off-line” LC/MALDI coupled MALDI-MS with liquid chromatography (LC) is used for a high sensitive measurement due to reducing an ionization suppression effect by separating the sample compounds. We have previously developed the high sensitive LC/MALDI “Advanced-nano-LC/MALDI-MS” system combined a nano-LC, a deposition probe integrated with a nano-LC column (column probe) and a DHB-prespotted focus plate to concentrate the droplets of nano-LC eluate. In this study, the advanced-nano-LC/MALDI-MS was applied to an analysis of E-cadherin glycoprotein from human plasma as a model sample. Our results indicated the identification of some N- and O-linked glycopeptides from a small volume of the plasma (10 μL). The O-linked glycopeptides of E-cadherin would be the first identification to our knowledge. These results demonstrate that the advanced-nano-LC/MALDI-MS system has great performance for an analysis of biological samples.
