MALDI-ISDにおけるシステイン含有タンパク質のN-Cα結合開裂とマトリックス効果
In-source decay (ISD) coupled with matrix-assisted laser desorption/ionization (MALDI) has been used for obtaining amino-acid sequence of proteins. The MALDI-ISD spectrum of cysteine-containing protein by using 5-amino-1-naphthol as a matrix showed intense ion peaks from the cleavage at the N-Cα bond of Xxx-Cys residue. In this study, we report the effect of matrix on the cleavage at the N-Cα bond of cysteine-containing protein backbone in MALDI-ISD.
